I don't know about 'best' but one sketch that I can watch every time and laugh is Chapelle's Knee-High Park (bootleg Sesame Street)
favorite scene: Stinky the Grouch (uncensored) whole episode: Knee-High Park (censored) hilarious

His DVD is fantastic, I only wish they hadn't cut some of the stuff out, like where he grinds down on Don Rickles even worse. His first dog show appearance was one of the funniest things I've ever seen. The guest who came out after asked Conan how he could do that to him, making him follow Triumph. I don't blame him. Anything's gonna look bad after that.
