Depends on what kind of trip, obv. I spent a lot of time on the road alone, and these songs struck a chord with me during that time:

James Taylor - Highway Song
Simon & Garfunkel - America, Homeward Bound

I think if you are thinking of "road trip" songs with a group of friends, you should be talking about whole albums. my personal road trip album favorites are

Bob Marley - Legend
Toadies - Rubberneck
Outkast - Aquemini
Clutch - Clutch

lol @ train, eve 6, counting crows. lolololol

most of my favorite road trip bands are just my favorite bands that don't put me to sleep. i.e. johnny cash, the pogues, tom waits, the clash, the holmes brothers, etc. pretty much any old blues works for me too - lightnin hopkins, howlin wolf, etc.

Yeah when I'm driving I just like it to be loud and fast

stuff like
The White Stripes - "Hotel Yorba"
Eagles of Death Metal - "I Want You So Hard (Boy's Bad News)"
Queens of the Stone Age - "You Think I Ain't Worth a Dollar, but I Feel Like a Millionaire"

etc. (just to pull a few things off recent "driving" mix CDs.

This is a good start but you need to add some Lynyrd Skynrd, Marshall Tucker Band and Allman Brothers.
