I don't know if they are profitable, but they generate revenue through banner advertising and custom pages like this one for NBC. I work in advertising and I probably have the advertising costs laying around somewhere on my desk. I'm guessing basic banner packages have to start at $1 million per month for just a small share of voice. YouTube is #4 in site traffic right now.
