AP .25/.50 - how much should I raise here?
 
Both players are about 25/10 over 40 hands or so.

How much should I RR here? I'm thinking $16-20 is the right amount, so I can get AI by the turn.

Probably $10-$12 It's not likely he's calling a 4bet with stacks that deep unless he has KK

But I do have to raise here though, right - because of SB being in the hand? Without the SB, can I play stop-n-go?

I doubt you will see a flop if you reraise $16-20.

I would make it $10. No need to go higher than $12 ever though.

Yikes you don't have to raise 16-20. Make it 12 and go from there.

We have no reason to assume they are competent.
I go for $12 at least, you want them to make a big mistake calling (and they might with any PP)
