I used to well up a lot, but the first movie that made tears fall was the Legend of Bagger Vance. The movie is a direct retelling of the Bhagavad Gita (Hindu spiritual text) using Georgia of the 1920s and golf. That means Will Smith, as Matt Damon's caddy/mentor, is essentially an incarnation of God.
So when it's time for Smith to leave he passes the torch to a little boy. The boy starts crying and says "what if something comes up and I don't know what to do?" and it gets me good.
I was pretty spiritual at the time so that probably factors into it a lot. The idea of God being right there physically and then leaving but saying he'll always be there in spirit or whatever is what did it.
