This may or may not qualify, but I throw out Kevin Costner in "No Way Out". I don't think anyone had heard of the dude prior to that but he had a lead role alongside Gene Hackman and (a dead) Sean Young. Pretty big breakout performance, and arguably his best, "Bull Durham" and "Dances With Wolves" being the others that come to mind.

This should not be a "breakout" movie that this person is known for - for example "Ed Burns in Brothers McMullen" does not qualify. Really, they should usually not even have a major part in the film.
